(HPLC: Organic Acid Analysis) When not using and rinsing the system

(HPLC: Organic Acid Analysis) When not using and rinsing the system
Category :

Answer

• When the analysis is complete, stop pumps as follows.
  1. Set the parameters as follows
    Pump A: 0.2 mL/min
    Pump B: 0.2 mL/min
    CTO: OFF
  2. Keep pumping with this setting until the separation column cools down to room temperature. and then, stop the operation of all pumps.
  3. Turn OFF each unit power.

• When not using the system for longer time
If you don’t perform analysis for 3 days or more, follow the additional procedures below:
Remove the columns. Be sure to seal the column inlet and outlet to store the columns. Attach the coupling 1.6C to the place where the columns were removed, and directly connect the autosampler outlet tubing to the reaction coil inlet tubing. Set water to the delivery pumps for eluent and buffer, and purge both pumps. Set flow rate of both pumps to 0.8 mL/min. Start pumping water to rinse the entire flow line for ten minutes. Turn OFF each unit power.

• Rinsing flow lines
After the installation of the Organic Acid Analysis System is complete, rinse out any contamination inside the flow lines by the procedure below before installing columns. If no analysis has been performed for a month or longer, you need to rinse the flow lines by the same procedure before analysis, because the flow lines may have become contaminated.

WARNING:
When diluting concentrated nitric acid, be sure to prepare water first and then slowly add concentrated nitric acid. Adding water to concentrated nitric acid may cause bumping due to the sudden generation of heat.
NEVER mix ethanol and high-concentrated nitric acid directly. An explosion may occur. When purging with these solutions in the same rinsing process, be sure to purge with purified water in between. Be careful not to mix even the waste liquid of those solutions after rinse operation.

CAUTION: In the procedure above, limit the time for rinsing with 1 mol/L nitric acid to about 10 minutes.
Do not pump longer than required. Otherwise, the plunger seal may deteriorate. Other solutions may be pumped longer without any problem.

NOTE:
Set the same rinse solution to two pumps and pump the solution from both pumps at 0.8 mL/min to rinse the entire flow line.
  No column is connected.

  1. Connect the autosampler outlet tubing and the plumbing case inlet tubing using a coupling 1.6C.
  2. Turn on power of eluent delivery pump, buffer delivery pump and autosampler.
  3. Purge eluent delivery pump and buffer delivery pump with ethanol, and wash the flow line for 10 minutes by delivering ethanol.
  4. Purge both pumps with water, and replace the solvent inside the flow line by delivering water for 5 minutes.
  5. Purge both pumps with 1 mol/L nitric acid, and deactivate the inside of flow line by delivering for 10 minutes.
  6. Purge both pumps three times with water, and replace the solvent inside the flow line by delivering water for 5 minutes.
  7. Purge both pumps with 10 mmol/L EDTA·2Na aqueous solution, and wash out the flow line for 30 minutes.
  8. Purge both pumps with water, and deliver for 60 minutes.
