« Analysis takes longer to complete »
• K is close to I and separation is poor.
→The column temperature is too low.: Check the column temperature.(40ºC)
• In general, the retention of amino acids is too great.
K is close to F.
→The specified column flow rate is too small.: Check the pump flow rate.(1mL/min)
• K, R, and H is particularly slow.
K is close to I and separation is poor.
R is close to ΔT and separation is poor.
H is close to G and separation is poor.
→The concentration of acetonitrile in the mobile phase is too low.: Add acetonitrile to the mobile phase to a concentration of 1% or replace the mobile phase with a new one.

• If fluctuations are observed in the retention of basic and acidic amino acids, it indicated that the pH, the salt concentration, or the ion-pair reagent concentration of the mobile phase has changed for some reason.: Replace the mobile phase with a new one.
* If a mobile phase is used for a long time, it is possible that the concentration of acetonitrile may decrease due to evaporation.
* Deterioration of the column can be identified from the degree of separation of the PTH-Met (M), Val (V), and Pro (P) peaks, and this can be used as an indication that the column should be replace. If the peaks have all broadened to a point where more than 50% of the peak height is overlapping, replace the column with a new one.
* If negative peaks are observed and analysis is affected, replace the mobile phase.
