(HPLC) SPD-M30A Handling and Cleaning of cell

(HPLC) SPD-M30A Handling and Cleaning of cell
Category :

Answer

<Cell Cleaning Method>
1. Flow 17 % phosphoric acid (5times dilution of 85 % phosphoric acid in water) for one hour at 1 mL/min.
2. Switch to water and flow for one hour at 1 mL/min.
3. Switch to methanol and flow for one hour at 1 mL/min.
4. Flow methanol or water at 1 mL/min while checking the baseline and continue until it is stable.
Note 1; The sensitivity may be decreased by the cleaning the flow cells with phosphoric acid in case that SPD-M30A is used as the front of LCMS or ELSD. We recommend you use 10% formic acid.
Note 2; The mobile phase which contains amine groups such as triethylamine tends to contaminate the cell capillary easily. So clean it once a month at least.

<Notes After Analysis>
Turn off the lamp or set the instrument to the sleep mode as soon as possible when the flow is stopped after an analysis finishes. The performance of cell may become low if the lamp continues to be lit while the mobile phases remain in the cell. The light intensity from the lamp to the cell can be restrained by setting the instrument to the sleep mode. In case of control by the LabSolutions, set “Shutdown” at the final line to turn off the lamp or let the instrument to sleep mode.

<Storage Method When Not Used for Long Periods>
When storing a cell with water and/or amine solvents remaining in it for a long period, the cell flow line sometimes becomes contaminated and the light intensity falls. Be sure to adopt one of the following methods when storing cells.
1. Flow methanol at once and remove the methanol by a syringe to let cells to be dry.
2. Fill the cells with 100% methanol, use a coupling to connect IN and OUT, and then exchange the methanol about once every month (rinsing for about 15 minutes).

<Other>
Purge the old mobile phase pooled in the flow line at the state that the flow cell is detached. To prevent the contaminants of the old mobile phase adheres to the flow cell.
When equilibrating after exchanging the column, please carry out the flow cell in a state separated from the flow path. To prevent the contaminants of the old mobile phase adheres to the flow cell.
