Points for Auto-Tuning necessity (LCMS)

Points for Auto-Tuning necessity (LCMS)
Category :

Answer

Auto-tuning is often implemented simultaneously in the following maintenance. There is a case that decreased sensitivity is solved only by auto-tuning. In auto-tuning, the resolution is automatically adjusted to approximately LCMS-8030/8040: 0.6, LCMS-8050/8060: 0.7 under the tuning conditions. The smaller than this value, the lower the signal intensity will become. (Alternatively, the greater the value gets, the higher the signal intensity will be). In these cases, it can be solved only by auto-tuning.

«Confirmation»
During the decrease in sensitivity, confirm the peak width on the manual tuning screen.
If the peak width is narrow, the signal intensity of chromatogram at the time of analysis also will decrease. The decrease in sensitivity is seen particularly in the area of m / z (molecular weight) of the peripheral target peak width. If the peak width is narrow in the entire area, sensitivity will be decreased regardless of component.

«Immediately after auto-tuning»
The full width at half maximum (FWHM) is adjusted to approximately 0.7.
«In the case of decreased sensitivity»
The full width at half maximum (FWHM) is lower than approximately 0.4, and the peak width becomes narrow.

«Corrective Action»
Implement the auto-tuning.
Be sure to implement auto-tuning after the degree of vacuum is fully raised. (Approximately for 6 to 12 hours).
