Replacing the Tubing
Answer
A high voltage (up to 5kV) is applied to the sample solution sprayed from the tip of the capillary ASSY in order to ionize the sample. Since a very fine-gauge tubing is used in the capillary ASSY, if any very small pieces of dirt get into the mobile phase or the sample, they can cause a blockage which will lead to a rise in the pump pressure.
If this happens, replace the tubing of the capillary ASSY.
If this happens, replace the tubing of the capillary ASSY.

1. Turn the LC pump OFF and disconnect the tubing from the LC.
2. Remove the ESI unit from the instrument.

3. Remove the ESI coupling.
a) Open the ESI cover(1).
b) Loosen the male nut(2) by hand and remove it.
c) Loosen the ESI coupling(3) with the spanner.
d) Remove the knurled screw(4).
a) Open the ESI cover(1).
b) Loosen the male nut(2) by hand and remove it.
c) Loosen the ESI coupling(3) with the spanner.
d) Remove the knurled screw(4).

e) Loosen the ESI coupling(5) completely, and remove it in the direction indicated by the arrow.

4. Replace the capillary ASSY.
a) Remove the capillary ASSY(2) from the ESI coupling(1).
b) Mount the new capillary ASSY(3) on the ESI coupling(1).
Tighten it firmly by hand.
a) Remove the capillary ASSY(2) from the ESI coupling(1).
b) Mount the new capillary ASSY(3) on the ESI coupling(1).
Tighten it firmly by hand.

5. Mount the ESI coupling in the ESI probe unit.
a) Insert the ESI coupling(1) into the ESI probe unit.

HInt: If the O-ring of the ESI coupling is damaged, replace it.
b) Tighten the ESI coupling(1) by hand and then turn it with a spanner, and adjust the projection of the tip.
Adjust so that the tip of the capillary ASSY projects about 0.5 to 1 mm.

6. Secure the ESI coupling in place.
a) Turn the knurled screw(1) by hand and fix the position of the ESI coupling.
b) Turn the male nut(2) by hand to secure the PEEK tube.
When securing the male nut(2), press against the PEEK tube(3) while tightening it.
a) Turn the knurled screw(1) by hand and fix the position of the ESI coupling.
b) Turn the male nut(2) by hand to secure the PEEK tube.
When securing the male nut(2), press against the PEEK tube(3) while tightening it.
