Answer

If a sample with a high salt content is analyzed with the protein sequencer, the background level in the chromatograms obtained may be high or the mobile phase may be contaminated. In this case, the sample can be desalted simply with the following procedure before performing sequence analysis.
 
1. Immerse a PVDF membrane cut to a size that fits into the PPSQ reactor in methanol.
 
2. Set the PVDF membrane in the reactor and load the sample.
  
 
3. Dry the sample (for approximately 10 minutes).
 
4. Immerse the PVDF membrane with the dried sample in 10 mL of 10% ethanol solution and apply ultrasonic treatment for 2 to 3 minutes to desalt the sample.
  If ultrasonic treatment is not possible, hold the PVDF membrane with tweezers, and rinse it in the 10% ethanol solution.
 
5. Take the PVDF membrane out of the 10% ethanol solution and dry it for 10 minutes in the reactor.

6 Set the reactor and start sequence analysis.
