(PPSQ) Pyridylethylation Sequence Schedule

Answer

The isocratic system allows pyridylethylation of cysteine in a PPSQ reactor preceding sequence analysis.
The sequence schedule for pyridylethylation is installed in the PPSQ.
 
NOTE: Pyridylethylation of cysteine in a PPSQ reactor is not recommended for the gradient system. Reductive alkylation is required as a pretreatment before sequence analysis.
 
1. Before starting sequence analysis, prepare the following reagent for pyridylethylation.
  0.16% 4 - vinylpyrizine
  0.08% tributyl phosphine
  in 80% acetonitrile
 
2. After adding a sample to a glass fiber disk that has been cleaned with polybrene, dry it in the usual way.
 
3. After drying the sample, add 10 μL of the pyridylethylation reagent specified above to the glass fiber disk.
 
4. Set the reactor in the sequencer without drying and start analysis immediately.
 
5. Click the [Schedule] button in the [PPSQ Analysis] window.
  
 
6. Click the combo box next to sequence schedule item that is displayed and select [PE Analysis] (pyridylethylation sequence schedule).
  
 
7. Regarding the other items, make the same settings used for the standard sequence schedule.
  HINT: This includes entering {number of analysis cycles + 1} as the number of cycles.
 
8. Click [start] button on the [Sequence] menu.
