(HPLC: Organic Acid Analysis) Precautions on Handling the Column

(HPLC: Organic Acid Analysis) Precautions on Handling the Column
Category :

Answer

Parts Number:
228-17893-91 Analysis Column,Shim-pack SCR-102H
228-17924-91 Guard Column SCR-102H (A guard column generally withstands approximately 500 times of use.)

• Precautions on Handling the Column
When the column is not used for seven days or more, seal it with the eluent, and store it in a cool, dark place.
Replace the eluent generally every three months.

• Column rinsing
Prevent column contamination by using a guard column.
Columns are consumables so the y are gradually deteriorated, resulting abnormal peak shapes and/or redaction of peak-resolutions In such cases, remove a guard column and check performance of an analytical columns alone by analyzing standard solution. If the result is good, change only the guard column to new one.
It may be possible to recover column performance by rinsing the column with following procedures.

1. Prepare 25 mmol/L of sulfuric acid aqueous solution for column rinsing
Mix 1.4mL of sulfuric acid (97% 18 mol/L) and 1L of water and set the obtained solvent and water to pump A and pump B respectively.
  WARNING:
Add sulfuric acid into water slowly and carefully. If water is added to sulfuric acid, there is a risk of sudden boiling due to sudden heat generation.
For your safety, make sure to put on guard glasses and gloves. If you touch sulfuric acid directly, Wash exposed part of your skin (especially eyes) immediately with water flow completely. The go to clinic.
2. Set “25 mmol / L sulfuric acid aqueous solution” for pump A and set water for pump B.
3. Connect wasting tube directly to column outlet. Rinsing the columns without plumbing case and detector.
HINT: Waste directly from the column.
4. Set parameters as follows,
  Pump A: 0.3 mL/min
  Pump B: 0 mL/min
  CTO: 80 ºC
5. Washing is performed overnight.
6. When washing is finished, place the tubes as their original state. The sulfate acid should be washed after disconnecting the column with water in the pump A.
7. Remove the column and connect the coupling 1.6C to the pipe.
8. Replace the sulfuric acid aqueous solution of pump A with water, and flow out the sulfuric acid solution in the piping by delivering the water.
